Humanin was first identified in 2001 from a cDNA library of the occipital lobe of an Alzheimer's disease patient as a factor that suppressed cell death induced by multiple familial AD-linked genes. This 24-amino acid peptide is encoded in the 16S rRNA region of mitochondrial DNA. Humanin signals through multiple pathways: it binds to IGFBP-3 to modulate IGF signaling, activates the STAT3 pathway through the trimeric CNTFR/WSX-1/gp130 receptor complex, and interacts with BAX to inhibit apoptosis. Humanin and its analogs have demonstrated protective effects against Alzheimer's, type 2 diabetes, atherosclerosis, and ischemia-reperfusion injury. It represents a key compound in mitochondrial biology and neuroprotection research.
